Antinociceptive effects of ethanolamides of palmitic and stearic acids and their composition (1:1) in rats with experimental arthritis

Abstract

Abstract. Modern methods of complex therapy of arthritis are aimed at controlling inflammation, but its often fail to cope with pain and can cause side effects.

N-acylethanolamines or acylethanolamides, such as palmitoylethanolamide and stearoylethanolamide, have analgesic and anti-inflammatory properties. The study is aimed at studying the antinociceptive effects of PEA, SEA and their combination with a single and course administration at a dose of 100 mg/kg in animals with experimental arthritis.

The aim to study the effect of single and course administration of palmitic and stearic fatty acid ethanolamides and compositions based on its in the implementation of antinociceptive effects in rats with experimental arthritis.

Methods. The studies were performed on white male Wistar rats (n = 60), divided into 10 groups. Arthritis was induced by intro-articular injection of Zymosan A from Saccharomyces cerevisiae (Sigma, USA; 4 mg/50 μl apyrogenic saline solution). The effects of fatty acid amides on nociceptive reactions in healthy rats and in rat with experimental arthritis were studied.

Results.A single administration of PEA, SEA, PEA: SEA (1:1) resulted in a significant reduction in nociceptive hyperreactions to mechanical and thermal stimuli in rats with Zymosan induced arthritis. The antinociceptive effects of the studied compounds were confirmed by recording the threshold of nociceptive reactions of the ipsilateral limb and the latent period of nociceptive reactions. Intragastric administration of course of РEA, SEA, PEA: SEA (1:1) to rats with experimental Zymosan-induced arthritis showed a more pronounced antinociceptive efficacy in relieve in ghyperalgesia to a mechanical stimulus compared to a single administration of the studied compounds.
